I use pagdefrag from sysinternals.
At startup it defragments system performance critical files that're locked when windows properly starts up like the pagefile or registry hives (these are those unmovable files defragger programs complain about). It takes ages the first time but now I have it all done every boot up in about 1-2 seconds.

Best defrager ever, for the desktop. But beware, this defrager is known to not be compatible with many games.

I use diskeeper, it runs pretty fast and calculates how much your read time can improve and it also has a boot-time defrag dealy, it's pretty sweet
